Nayudu was born on October 31, 1895, in Bara Bada Nagpur, to Cottari Surya Prakash Rao Nayudu, the son of Rao Bahadur Cottari Narayana Swamy Nayudu, a wealthy balija naidu from Machilipatnam, Andhra Pradesh, who was a lawyer and landowner with numerous villages and a large portion of Nagpur. He was a founding member of the All India National Congress party in addition to being a successful lawyer. Narayana Swamy has enough money to send both of his sons to England to pursue their education. Cottari Vekatramana Nayudu, his eldest son, married the daughter of Raja Prabhakar Moorthy of Eluru and had one son, Cottari Ranga Rao Nayudu, who was also an attorney in England.
